README.postfixを元に以下の手順で行ってみました。
#  ./configure --with-runtime-dir=/var/amavis --with-amavisuser=vscan 
--enable-postfix --enable-all

# make

# make install

/etc/postfix/main.cf
2行追加
content_filter = vscan:
soft_bounce = yes

/etc/postfix/master.cf
2行追加
vscan            unix  -  n  n  -  10  pipe user=amavis 
argv=/usr/sbin/amavis ${sender} ${recipient}localhost:10025  inet  n  -  n  
-  -   smtpd -o content_filter =

けれども、現象はまったく変わりませんでした・・・。
このルールを明記すると相変わらず、spoolに情報が溜まりません。

ログの内容は以下の通りです。

一応結果まで報告致しました。このエラーを元にもう少し調べようと思います。
